Retatrutide When Lean: Glucagon, Glycogen & Training
A viral X thread says retatrutide should be avoided under 20% body fat because glucagon agonism drains glycogen and kills the gym pump. The liver part is real physiology. The rest has never been tested.
Four things the glycogen argument gets right and wrong
Glucagon raises blood glucose by increasing liver glycogen breakdown (glycogenolysis) and glucose production (gluconeogenesis) while decreasing glycogen storage (glycogenesis) and glycolysis (PMID 12626323). In the liver it also increases fatty acid oxidation and helps regulate amino acid and ketone metabolism (PMID 38300523). So the thread is right that the glucagon arm of retatrutide pushes the liver away from storing glycogen.
The two stores behave differently. In a 13C MRS study of nine male cyclists doing high-intensity intervals, muscle glycogen was used preferentially, and carbohydrate feeding cut muscle glycogen use by about 40% while liver glycogen use was not significantly different (PMID 42552237). The pump is a muscle event. No retatrutide study has measured muscle glycogen, liver glycogen, or gym performance, so "this is why you cannot get a pump" is a hypothesis, not a finding.
A glucagon infusion raised energy expenditure by 15% in 11 healthy men, a rise similar to cold exposure, without activating brown fat (PMID 26434748). In Lilly's discovery work in obese mice, retatrutide combined GIPR- and GLP-1R-driven reductions in calorie intake with GCGR-driven increases in energy expenditure (PMID 35985340). A 2023 review flags glucagon-driven lowering of blood amino acids as an open issue for glucagon multi-agonists and notes the human mechanism is not fully understood (PMID 36736539).
In a DXA substudy of adults with type 2 diabetes (BMI 25 to 50), retatrutide 8 mg cut total fat mass by 26.1% at 36 weeks, and the share of weight lost as lean mass was similar to other obesity treatments (PMID 40609566). The phase 2 obesity trial enrolled adults with BMI 30 or higher, or 27 to 30 with a weight-related condition, and reported dose-dependent heart-rate increases that peaked at 24 weeks (PMID 37366315). None of this tells you what happens at 15% body fat.
What the Data Shows
Key Takeaways
- Glucagon shifts the liver toward glycogen breakdown and glucose output and away from glycogen storage (PMID 12626323).
- Liver and muscle glycogen are separate stores; during interval exercise muscle glycogen was used preferentially (PMID 42552237).
- Glucagon infusion raised energy expenditure by 15% in healthy men (PMID 26434748).
- Retatrutide trials enrolled adults with obesity, overweight plus a weight-related condition, or type 2 diabetes (PMIDs 37366315, 40609566, 42814954).
- In a DXA substudy, the share of weight lost as lean mass on retatrutide was similar to other obesity treatments (PMID 40609566).
- Orforglipron (LY3502970) is an orally active, non-peptide GLP-1 receptor agonist that was selective against other class B receptors in lab testing (PMID 33177239).
- Pairing exercise with a GLP-1 drug (liraglutide) improved body-fat percentage and cardiorespiratory fitness more than either alone (PMID 33951361).
- Whether retatrutide lowers muscle glycogen, the pump, strength, or training performance. No study has measured these.
- How retatrutide behaves in lean, resistance-trained adults. The 20% body-fat cutoff in the thread has no trial behind it.
- Whether glucagon-driven lowering of blood amino acids causes more muscle loss in lean people (PMID 36736539).
- Whether tirzepatide, orforglipron, or exenatide is better for lean lifters. There are no head-to-head trials in athletes.
- How UBT251 compares with retatrutide. We found no peer-reviewed head-to-head data.
Frequently Asked Questions
Does retatrutide kill your pump in the gym?
Nobody has tested it. Glucagon does push the liver toward glycogen breakdown and glucose output (PMID 12626323), but the pump happens in muscle, and liver and muscle glycogen are separate stores that behave differently during exercise (PMID 42552237). No retatrutide trial measured muscle glycogen or gym performance. A drop in food intake is a simpler explanation worth ruling out first.
Is retatrutide a bad idea if you are already lean?
There is no data either way. The phase 2 and phase 3 trials enrolled adults with obesity, or overweight with a weight-related condition, and the body-composition substudy enrolled adults with type 2 diabetes. The "under 20% body fat" cutoff in the viral thread is an opinion, not a trial result. Retatrutide is also not an approved drug (PMIDs 37366315, 40609566, 42814954).
Does glucagon agonism deplete muscle glycogen?
The well-described glycogen effects of glucagon are in the liver: it increases glycogen breakdown and glucose output and decreases glycogen storage there. No retatrutide study has measured muscle glycogen. In cyclists, carbohydrate feeding spared muscle glycogen during intervals but did not significantly change liver glycogen use, which shows the two stores do not move together (PMIDs 12626323, 42552237).
Does retatrutide cause muscle loss?
It causes some lean-mass loss along with fat loss, like other weight-loss drugs. In a DXA substudy of adults with type 2 diabetes, fat mass fell 26.1% on 8 mg over 36 weeks, and the share of weight lost as lean mass was similar to other obesity treatments. Lean, trained people were not studied (PMID 40609566).
Can retatrutide be detected in an athlete drug test?
A 2026 study built a plasma test for retatrutide to WADA laboratory criteria. In four users who self-injected 2 mg on days 1 and 7, retatrutide stayed detectable in plasma for about 56 days after the first dose, but it was not found in urine. The authors note growing off-label use among physically active people (PMID 42745421).
